Indication & Clinical Use

Semaglutide is indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus. It is also indicated for chronic weight management in adults with obesity or overweight with at least one weight-related comorbidity.

Dosing & Administration

Semaglutide is typically started at a low dose, such as 0.25 mg once weekly. The dose is then gradually increased over several weeks under medical supervision. This titration helps reduce potential gastrointestinal side effects. The maximum dose is determined by your healthcare provider.

What It Is

Semaglutide is a medication used to help manage blood sugar levels in adults with type 2 diabetes. It is also used to support weight management in certain individuals. It is administered as a once-weekly injection.

How It Works

Semaglutide works by mimicking a natural hormone in your body called GLP-1. This hormone helps regulate blood sugar by increasing insulin release when blood sugar is high and decreasing glucagon secretion. It also slows gastric emptying and can reduce appetite, which contributes to weight management.

Benefits

Using Semaglutide can lead to improved blood sugar control for individuals with type 2 diabetes. For those using it for weight management, it can help reduce body weight. Some formulations of semaglutide have also been shown to reduce the risk of major adverse cardiovascular events in adults with type 2 diabetes and established cardiovascular disease.

Important Safety Information

Serious Warnings & Precautions

Semaglutide has caused thyroid C-cell tumors in rodents. It is not known if it causes these tumors, including medullary thyroid carcinoma (MTC), in humans. Pancreatitis has been reported in patients treated with GLP-1 receptor agonists. Monitor for symptoms of pancreatitis. Acute gallbladder disease, including cholelithiasis or cholecystitis, has been reported. Monitor for signs and symptoms. Renal impairment has been observed in patients treated with GLP-1 receptor agonists. Monitor kidney function in patients with renal impairment.

Contraindications

Do not use Semaglutide if you or your family have a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Do not use if you have a known hypersensitivity to semaglutide or any of its components. It is not indicated for use in patients with type 1 diabetes.

Common Side Effects

Common side effects may include nausea, vomiting, diarrhea, abdominal pain, and constipation. These side effects are often mild and tend to decrease over time as your body adjusts to the medication. Following the recommended dosing schedule can help manage these effects.

Drug Interactions

Semaglutide may affect the absorption of orally administered medications due to slowed gastric emptying. Use with caution with other medications that lower blood sugar, such as insulin or sulfonylureas, as this may increase the risk of hypoglycemia. Your healthcare provider will review your current medications.

Important Safety Information

Always discuss your full medical history and all medications you are taking with your healthcare provider before starting Semaglutide. Report any unusual symptoms, especially severe abdominal pain or signs of an allergic reaction. Do not share your injection pen with others, even if the needle is changed, as this carries a risk of infection.
